There are no doors in Jefferson that are "special game locked" doors. There are no characters in that game that you can kill that will result in the game ending prematurely. Tom Hall made one of my favrouite RPGs of all time, Anachronox - if Midway would fund a project like that for him and you to work on...... sigh.....
