like quests in act one can be gone after you enter act 2 and quest from act 2 are blocked after you get to act 3 and so on?

Not that I've noticed.

I found that I auto-failed a quest I hadn't picked up yet after moving to act III. So I would say yes, some quests can become unavailable/unable to finish after you move to another act.

Not really a good topic for non-spoilers forum..

I know that a certain wizard companion's  personal quest has a moment where it has to be advanced in act II and before going to act III, as

 

 

 

Certain location needed to be visited is in Defiance Bay

For the most part it's just temporary, there a couple quests you lost out on if not finished before the end of Act II but the rest you only lose temporary access to.
